Dose-dependent biological damage of tumour cells by laser-accelerated proton beams

چکیده
We report on the first irradiation of in vitro tumour cells with laser-accelerated proton pulses showing dose-dependent biological damage. This experiment, paving the way for future radiobiological studies with laser-accelerated protons, demonstrates the simultaneous availability of all the components indispensable for systematic radiobiological studies: a laser–plasma accelerator providing proton spectra with maximum energy exceeding 15MeV and applicable doses of a few Gy within a few minutes; a beam transport and filtering system; an in-air irradiation site; and a dosimetry system providing both online dose monitoring and absolute dose information applied to the cell sample and the full infrastructure for analysing radiation-induced damage in cells. 4 Authors to whom any correspondence should be addressed. Dose estimation with the calibration of dose-response curve of micronucleus in human peripheral lymphocytes induced by 50MeV proton beams
Background: The purpose of this paper is to establish an easy and reliable biodosimeter protocol to evaluate the biological effects of proton beams. Materials and Methods: Human peripheral blood lymphocytes were irradiated using proton beams (LET: 34.6 keV μm−1), and the chromosome aberrations induced were analyzed using cytokinesis-blocked (CB) micronucleus (MN) assay. To determine th...
متن کاملAnalysis of Relative Biological Effectiveness of Proton Beams and Iso-effective Dose Profiles Using Geant4
Background: The assessment of RBE quantity in the treatment of cancer tumors with proton beams in treatment planning systems (TPS) is of high significance. Given the significance of the issue and the studies conducted in the literature, this quantity is fixed and is taken as equal to 1.1.Objective: The main objective of this study was to assess RBE quantity of proton beams and their variations ...
متن کاملUltra-short laser-accelerated proton pulses have similar DNA-damaging effectiveness but produce less immediate nitroxidative stress than conventional proton beams
Ultra-short proton pulses originating from laser-plasma accelerators can provide instantaneous dose rates at least 10(7)-fold in excess of conventional, continuous proton beams. The impact of such extremely high proton dose rates on A549 human lung cancer cells was compared with conventionally accelerated protons and 90 keV X-rays. Between 0.2 and 2 Gy, the yield of DNA double strand breaks (fo...
متن کاملTumour radiobiology beyond fractionation
Historically it has been shown repeatedly that single high doses of radiation do not allow a therapeutic differential between tumor and critical normal tissues but dose fractionation does. The purpose of conventional dose fractionation is to increase dose to the tumor while preserving normal tissue function. Tumors are generally irradiated with 2Gy dose per fraction delivered daily to a more or...
متن کاملEvaluation of variable relative biological effectiveness and the creation of homogenous biological dose in the tumor region in helium ion radiation to the V79 cell line
In radiation therapy, ions heavier than proton have more biological advantages than a proton beam. Recently, ion helium has been considered due to high linear energy transfer (LET) to the medium and a higher relative biological effect (RBE). To design the spread-out Bragg peak (SOBP) of biological dose for radiation with any type of ion, we need exact values of RBE, which is dependent to dose, ...
